Frequently Asked Questions About Submitting Libraries for Sequencing

What quantity of pooled library do we need for sequencing service?
We request that at least 1 µg of pooled library be submitted for sequencing service at a concentration of at least 20 ng/ µL. Although 500 ng total library is typically enough, we prefer to have at least 1 µg for a possible repeat experiment. If you do not choose us for sequencing, please refer to your sequencing vendor for their requirements.
What are the sample requirements for shipping a pre-prepared library sample for sequencing (buffer, tubes, etc)?
Buffer: Pre-prepared libraries may be suspended in a wide variety of buffers, including 10 mM Tris, dH2O, or Qiagen EB. The most critical requirement is that buffers contain no more than 0.1 mM EDTA.
Tubes: All samples should be submitted in 1.5 mL centrifuge tubes. Please include the identifying information of the sample on both the top and the side of the tube such as sample number, sample name, concentration, and quantity, matching the sample submission form.
