iRepertoire offers multiplex Primer mixes, Amplification Services, and Sequencing Services...

 

To facilitate your research, we have developed mPCR primer-mix product lines for V(D)J amplification and sequencing. We offer Primers, Amplification Services, and Sequencing Services for both Human and Mouse samples; T and B cells, including all the chains; and we have primers for both the Roche 454 and the Illumina sequencing platforms. We also have at least 10 barcodes per reagent system, so that multiple samples can be pooled into one sequencing lane to reduce the cost associated with sequencing.

 

Samples can be peripheral blood, sorted T or B cells, or tissues.  The majority of our primer systems are compatible with RNA only; however, we do offer human TCR beta Illumina primers compatible with both gDNA (V-J primers) and RNA (V-J and VC primers). Our primers can utilize several sequencing platforms, including the Illumina platform with 100 paired-end reads (100-PER; GA IIx, HiSeq2000, and MiSeq), the Illumina platform with 250-paired-end reads (250-PER; MiSeq), and Roche 454.

 

 

If you are interested in T cell studies...

 

For T cell studies, we offer V-J (Human TCR beta) and V-C primers (all of the chains) for the Illumina platform with 100-PERs (GA IIx, HiSeq2000, and MiSeq). For this platform, the V-J primers amplify about 100 bp sequences around the CDR3 region and are suitable for both genomic DNA and RNA samples; the V-C primers amplify about 150 bp sequences around the CDR3 region and are appropriate for RNA samples only because there is a large intron in between the C and rearranged V(D)J genes. For the Illumina MiSeq platform with 250-PERs and Roche 454, we offer V-C primers. For this platform, the primers amplify about 450 bp of the sequence around the CDR3 region and are suitable RNA samples only.  We are now offering an amplification-to-sequencing serivce on the Illumina MiSeq for $650 per sample. If you are interested in B cell studies...

 

For B cell studies, we have V-C gene primers for all of the Illumina platforms and Roche 454 platforms. We recommend using the Illumina 250-PER MiSeq primers or the Roche 454 primers for B cell repertoire amplification, if possible. Both the MiSeq and Roche 454 primers allow for a longer read length, and a large portion of antibody sequence information can be obtained beginning from within the first framework region and extending into the beginning of the C-region. The sequence output length will be the same for both Roche 454 and Illumina MiSeq; however, MiSeq offers the option of deeper sequencing coverage (ie, more sequencing reads) at a lower sequencing cost. We are now offering an amplification-to-sequencing serivce on the Illumina MiSeq for $650 per sample.
